TRDizin Antioxidant Enzyme Activities of Some Wild and Cultivated Edible Mushrooms in Turkey(2020-08-01) Şeyma Selin Akın; Aysun Pekşen; Sezgin Ayan; Nezahat TurfanIn this study; wild and cultivated edible mushrooms [Boletus edulisBull.:Fr, Craterellus cornucopioides(L.) Pers., Lactarius deliciosus(L.ex Fr.) S.F.Gray, Laetiporus sulphureus(Bull.:Fr.) Murr., Marasmius oreades(Bolt. ex Fr.) Fr., Morchella conicaPers., Ramaria botrytis(Pers.:Fr.) Ricken, Tricholoma terreum(Schaeff.: Fr.) P.Kumm., Hericium erinaceus(Bull.:Fr.) Pers., Lentinula edodes(Berk.) Pegler, Ganoderma lucidum(Curt.:Fr.) P. Karst., and Pleurotus ostreatus(Jacq. ex Fr.) P. Kumm. (1-4)] were obtained from different locations in Turkey. Phenylalanine ammonia lyase (PAL) enzyme activity, ascorbate peroxidase (APX), peroxidase (POD), and superoxide dismutase (SOD) activity changesand nitrate, β-carotene and lycopene levels were investigated in 15 samples to determine antioxidant enzyme capacity. As a result of the study, the highest amount of β-carotene and lycopene were determined in H. erinaceus.P. ostreatus-2 had the lowest amount of β-carotene, whereas Pleurotus ostreatus-1 had the lowest amount of lycopene. Species rich in nitrate content wereC. cornucopioidesand P. ostreatus-4. P. ostreatus-3 wasthe poorest species in terms of nitrate compared to other mushroom samples. PAL activity of mushrooms varied between 5.863 and 8.893 EU mg-1protein. For APX values, P. ostreatus-4 had the highest value, while H. erinaceusspecies had the lowest value. Among mushroom species, the highest and the lowest POD values were determined in H. erinaceusand B. edulis, respectively. C. cornucopioideshad the highest and P. ostreatus-3had the lowest SOD values.TRDizin Assessment of The Effects of Silicon Application on Growth Parameters and Some Bioactive Components in Hungary vetch (Vicia pannonica Crantz)(2022-01-01) Nezahat TurfanThe objective of this study was to investigate the effect of 10 mM silicon (Si) application on salt stress (50, 100, and 200 mM NaCl) tolerance in Hungarian vetch seedlings (Ege Beyazi-79) based on growth parameters (shoot length and fresh weight, organic dry matter, and moisture), bioactive compounds as chlorophyll, carotenoid, proline, protein, nitrate, and nutrient status. Silicon was applied to the seedlings from the leaves and NaCl from the salt soil, both of which were dissolved in Hoagland solution. The results revealed that the highest shoot length Si+100 mM NaCl was recorded and the highest fresh and dry weight was recorded with length Si+50 mM NaCl. The highest dry matter was obtained from Si and 200 mM NaCl. The highest chlorophyll, carotenoid, proline, protein, and nitrate were obtained from 100 mM NaCl+Si, while the lowest chlorophyll, carotenoid, and nitrate were obtained from 200 mm NaCl and the lowest proline and protein from 100 mM NaCl, respectively. The amount of K, Mg, and S were low in NaCl doses but high in Si and Si + NaCl doses, and P was higher in all groups compared to the control. The content of Na, Co, and Se were high in all groups except Cl 50 NaCl, while Mn, Zn, and Ni were higher in Si and Si + NaCl doses. Silicon content of samples was high at higher NaCl + Si doses. For this purpose, the measurement of yield was performed with bulbs,but the amount of some bioactive chemicals as chlorophyll, carotenoid, flavonoid, phenolic, proline,protein, free amino acid, sugars, pyruvic acid, malondialdehyde (MDA), hydrogen peroxide (H2O2),ascorbate peroxidase (APX) and superoxide dismutase (SOD) activities obtained from the fresh leaf andcloves grown in the different soil mix. According to the result, while the amount of pigment was foundto be higher in the leaf of FS and FS-IPT, the flavonoid in FS-CHICKEN and total phenolic in FS-CATTLEGOAT samples, nitrogenous compounds, and sugars (glucose, sucrose) were found to be rich in thecontrol group (open field). The measurement of bulb weight, length, diameter, and the amount oflycopene, flavonoid, total phenolic, and free amino acid of garlic cloves obtained from FS-CHICKEN-IPT,FS-CATTLE-GOAT, FS-CHICKEN-GOAT, and FS-GOAT-NPT was higher when compared to the control.Furthermore, APX and SOD increased, whereas malondialdehyde lowered in these samples. As a result,the amounts of bulb yield, enzymatic and non-enzymatic compound, and sugars estimated usingorganic fertilizer were higher than the control, which was grown with inorganic fertilizer in an open field.In addition, the most drought-susceptible examples are the samples of FS, FS-ITP, FS-NTP, and FSCATTLE soils considering all data.TRDizin Variation in Chemical Constituents of Siyez Wheat (Triticum monococcum L.) in Response to Some Abiotic Stress Factors(2019-01-01) Temel Sarıyıldız; Ekrem Mutlu; Nezahat TurfanMain aim of this study was to determine the effects of different salt contents (75 mM, 150 mMand 225 mM NaCl), heavy metal (0.2 mg/L FeCl3, NiCl2, ZnCl2), lime (2 mg/L CaCO3), drought(50%) and pollution (0.2 mg/L dust of factories) on photosynthetic pigments, malondialdehyde(MDA), hydrogen peroxide (H2O2) levels, the ascorbate peroxidase (APX), catalase (CAT),guaiacol peroxidase (GPOX) and superoxide dismutase (SOD) in Siyez wheat (Triticummonococcum L.). All experiments were carried out under laboratory conditions with 16 hour-dayand 8 hour-night photoperiod in an incubator at 23 ± 1°C. Results showed that mean chlorophylla concentration was highest in the siyez seedlings treated with the pollution, while both meanchlorophyll-b and total chlorophyll concentrations were highest with 75 mM salt application.Mean total carotenoid was, however, highest with the drought treatment and mean relative watercontent was highest with NiCl2 application. Mean MDA and H2O2 contents were found to behighest in the siyez seedlings treated with 225 mM salt, whereas they were lowest with NiCl2treatment. Mean proline content was highest with the NiCl2 treatment compared to the lowestconcentration in the control siyez seedlings (82 µmol/g). Mean APX, CAT and GPOX activitieswere noted to be highest in the siyez seedlings treated with NiCl2. In general, the siyez seedlingsshowed high tolerance to the pollution, NiCl2 and drought with having highest photosyntheticpigments, proline, protein content and enzymes activities. Among all treatments, 225 mM NaCland CaCO3 negatively influenced chemical compounds of the siyez seedling. When all data aretaken into consideration, it can be said that higher photosynthetic pigments, proline contents,antioxidant enzymes activities and lower MDA and H2O2 levels play an important role in theresistance of siyez seedlings against abiotic stress conditions.
