Item Ayçiçeği bitkisinde ısı şoku proteini ve ısı şoku faktörü genlerinin özelliklerinin incelenmesi(Fen Bilimleri Enstitüsü, 2022-07) Ceylan, YusufIsı şoku proteinleri (Hsp) canlıların gelişimini ve canlılığını sürdürebilmesi için hayati öneme sahip protein ailesidir. Yüksek sıcaklık, tuzluluk, kuraklık gibi abiyotik stres koşulları karşısında bu proteinlerin ekspresyon seviyeleri artmaktadır. Bu çalışmada ayçiçeği bitkisindeki ısı şoku proteinleri (Hsp) ve ısı şoku faktötleri (HSF) gen aileleri biyoinformatik analizler ile belirlenmiş ve incelenmiştir. Ayrıca belirlenen bu genlerin, ayçiçeği çeşitlerinde yüksek sıcaklık, kuraklık ve yüksek sıcaklık-kuraklık kombine stres koşulları altında gen ifade analizleri yapılmıştır. Ayçiçeği genomunda HSF ailesine ait 88 gen, sHsp gen ailesine ait 72 gen, Hsp40 ailesine ait 192 gen, Hsp60 ailesine ait 52 gen, Hsp70 ailesine ait 85 gen, Hsp90 ailesine ait 49 gen ve Hsp100 ailesine ait 148 gen tespit ve analiz edilmiştir. Proteinlere ait motif yapılarının aynı filogenetik ağaçta yer alan proteinler için benzer olduğu belirlenmiştir. Proteinlere ait üç boyutlu yapı tahminlemesinde HansHsp hariç, tamamında α-sarmal yapının baskın olduğu gözlenmiştir. Gen ontoloji analizinde Hsp genlerinin çoğunlukla bağlanma moleküler fonksiyonuna sahip olduğu; hücresel ve metabolik süreçlerde görev aldığı ve hücrenin sitozol ve organeller olmak üzere birçok bölgesinde yer aldığı saptanmıştır. Yüksek sıcaklık, kuraklık ve yüksek sıcaklık-kuraklık kombine stres koşullarında 0.; 0,5.; 1. ve 2. saatlerde toplanan yaprak örneklerinin gen ifade seviyelerine gerçek zamanlı PZR ile bakılmıştır. Stres koşullarında genellikle ilk yarım ve birinci saatlerde gen ifadelerinde artış görülmüştür. HanHSF-45 ve HanHsp70-29 genleri ifadelerinin, hem yüksek sıcaklık hem de yüksek sıcaklık-kuraklık kombine stres koşullarında çalışılan her iki ayçiçeği çeşidinde de artış gösterdiği ve bu streslere karşı potansiyel markör genler olabileceği düşünülmektedir. Çalışmamız, hayati bir öneme sahip olan ısı şoku faktörü ve ısı şoku proteinlerinin yapısının, organizasyonunun, evriminin ve gen ifade analizlerinin anlaşılması açısından önem arz etmektedir. Heat shock proteins (Hsps) are protein families vital for all organisms' growth and survival. Besides the ordinary conditions, the expression of these proteins ascends during abiotic stress factors such as high temperature, salinity, and drought. Using bioinformatics approaches, the current study identified and analyzed HSF and Hsp gene family members in the sunflower (Helianthus annuus L.) plant. HSF, sHsp, Hsp40, Hsp60, Hsp70, Hsp90, and Hsp100 domains were analyzed in the sunflower genome, and 88, 72, 192, 52, 85, 49, and 148 genes were identified, respectively. The motif structures of the proteins in the same phylogenetic tree were similar, and the α-helical form was dominant in all the protein families except for sHsp. The estimated three-dimensional structure of 28 sHsp proteins was determined as β-sheets. Considering protein-protein interactions, the Hsp60-09 protein (38 interactions) was found to be the most interacting protein. The most orthologous gene pairs (58 genes) were identified between Hsp70 genes and Arabidopsis genes. The expression analysis of selected genes was performed under high temperature, drought, and high temperature-drought combined stress conditions in two sunflower cultivars. In stress conditions, gene expressions were upregulated for almost all genes in the first half and first hours at large. The expressions of HanHSF-45 and HanHsp70-29 genes were raised in two cultivars under high temperature and high temperature-drought combined stress conditions. This study presents a blueprint for subsequent research and delivers comprehensive knowledge of this vital protein domain.
