TRDizin Effect of Tocilizumab on Acinetobacter baumannii Lung Infection in an Immunosuppressed Rat Model(2022-02-01) Ozgur CELEBI; Irfan CINAR; Serkan YILDIRIM; Kagan Tolga CINISLI; Demet CELEBI; Iclal HALICI; Zekai HALICI; Nurullah AKGUN; Halil KESKINOur study aimed to investigate effect of tocilizumab on the lung tissue in the presence of Acinetobacter baumannii infection in immunosuppressed rats. A forty-eight female Wistar albino rats were divided equally into eight groups: Group 1: Healthy (H), Group 2: Immunosuppressed (IM), Group 3: Healthy rats given A. baumannii bacteria (H+BAC), Group 4: Immunosuppressed rats given A. baumannii bacteria (IM+BAC), Group 5: Healthy rats given tocilizumab (H+TCZ), Group 6: Immunosuppressed rats given tocilizumab (IM+TCZ), Group 7: Healthy rats given A. baumannii bacteria and tocilizumab (H+BAC+TCZ), Group 8: Immunosuppressed rats given tocilizumab and A. baumannii bacteria (IM+BAC+TCZ). Fourteen days after the immunosuppression of group 2, 4, 6 and 8 with hydrocortisone, group 3, 4, 7 and 8 were A. baumannii was dropped into the trachea. One hour after A. baumannii application, TCZ was administered to Groups 5, 6, 7 and 8. NF-κB, IL-6 and NLRP3 mRNA expressions were decreased in the IM group compared to the healthy group (P<0.05). Although NF-κB, IL-6 and NLRP3 mRNA expression decreased in the IM+TCZ group compared to the healthy group (P<0.05) NF-κB, IL-6 and NLRP3 mRNA expression increased in the H+TCZ group (P<0.05). Despite decreasing cytokines, A. baumannii has been shown to increase infection-related lung injury. This suggests that in patients currently or recently using steroids, tocilizumab may increase organ damage due to opportunistic infection.TRDizin Efficacy of boric acid therapy in a pseudomonas aeruginosa-induced chronic otitis media model in rats(2022-09-30) Muhammed Sedat SAKAT; Ayhan KARS; Abdulkadir ŞAHİN; Serkan YILDIRIM; Korhan KILIÇ; Sinan KÖYCEĞİZPurpose: Chronic otitis media (COM) is one of the common infectious diseases of the middle ear caused by bacterial or viral pathogens. The purpose of the present study was to investigate the efficacy of boric acid (BA) in the treatment of COM by comparing topical 4% and 8% BA applications with systemic ciprofloxacin in a Pseudomonas aeruginosa (P. aeruginosa) inoculation-induced model of COM in rats. Materials and Methods: Forty-two Sprague Dawley rats were divided into seven equal groups. The COM model was created with P. aeruginosa. Clinical, histopathological and, immunohistochemical comparisons were performed between the control, COM, topical 4% and 8% BA treatment, a systemic ciprofloxacin treatment, and topical 4% and 8% BA contact groups. Results: In the COM model, moderate edema, inflammation, degeneration and moderate tumor necrosis factor-alpha (TNF-α) expression were detected with the application of 4% BA. Mild edema, inflammation, degeneration, and mild TNF-α expression were detected with 8% BA application. Conclusion: Both 4% and 8% BA treatment provided significant clinical, histopathological and immunohistochemical improvement. The application of 8% BA was found to exhibit greater therapeutic efficacy, with no harmful effect on the middle ear mucosa.TRDizin Oleanolik asidin çekal ligasyon ve punksiyon ile sepsisoluşturulan ratlarda kalp hasarı üzerine etkileri(2020-12-01) Tuba AYDIN; Mesut Bünyami HALICI; Kübra COŞAR; Hüseyin Serkan EROL; Mohammed ALHİLAL; Serkan YILDIRIM; Muhammet Bahaeddin DÖRTBUDAKBu çalışmada zeytin ağacı (Olea europeae) ağacının yaprağından saflaştırılan oleanolik asidin (OEA) ratlarda çekal ligasyon ve punksiyonla (CLP) indüklenen sepisisin oluşturduğu kalp hasarı üzerine etkileri incelendi. Her gruba eşit olacak şekilde 30 adet Wistar erkek rat sham, sepsis, OEA-150, OEA-300 ve CEFT olarak 5 gruba ayrıldı. Kromatografik metotlarla zeytin yaprağından saflaştırılan oleanolik asit OEA gruplarına 150 ve 300 mg/kg dozlarda oral yolla uygulandı. CEFT grubuna seftriakson 150 mg/kg dozda intraperitoneal yolla uygulandı. Uygulamadan 24 saat sonra ratlardan kan serumu ve kalp dokuları, histopatolojik, immünohistokimyasal ve biyokimyasal analizler için alındı. Yapılan histopatolojik ve immünohistokimyasal incelemede, sepsis grubunun doku tümör nekrozis faktör (TNF)-a ve interlökin (IL)-1β ekspresyonlarında artış, myokartta kas liflerinde şiddetli düzeyde hyalin dejenerasyon, Zenker nekrozu ve damarlarda şiddetli düzeyde hiperemi gözlendi. OEA ve seftriakson (CEFT) sitokin ekspresyonlarında ve histopatolojik bulgularda azalma sağladı. Sepsis, doku superoksit dismutaz aktivitesini (SOD) azalttı. Ayrıca lipid peroksidasyonu (LPO) ve glutatyon (GSH) seviyeleri ile doku katalaz (KAT), serum kreatin kinaz (CK-MB) ve serum laktat dehidrojenaz (LDH) aktivitelerini önemli şekilde arttırdı. OEA ve CEFT, SOD aktivitesini önemli derecede arttırırken diğer parametrelerde belirgin düşüşe neden oldu. Bu çalışmadan elde edilen bulgular ışığında oleanolik asidin sepsis sırasında oluşan kalp dokusu hasarının azaltılmasında faydalı olabileceği düşünülmektedir.TRDizin The effects of oleuropein on lung and heart injury in cecal ligation and puncture - induced sepsis(2020-03-01) Tuba AYDIN; Hüseyin Serkan EROL; Serkan YILDIRIM; Mesut Bünyami HALICI; İsmail CANAim: Sepsis is a fatal serious clinical disorder. The oxidative stressin sepsis plays an important role in the inflammation. Therefore, theeffects of oleuropein (OLE) and ceftriaxone (CEFT) on sepsis wereinvestigated in this study.Materials and Methods: Thirty male rats were divided into fiveequal groups: Sham, Sepsis, CEFT, OLE-150 and OLE-300, The150 mg/kg of ceftriaxone (i.p.), and 150 and 300 mg/kg OLE (p.o.)were administered to the treatment groups fifteen minutes beforethe experiment. After 24 hours, blood samples, lungs and hearts tissues were taken. The creatine kinase (CK) and lactate dehydrogenase (LDH) activities in serum, lipid peroxidation (LPO), glutathione(GSH), superoxide dismutase (SOD) and catalase (CAT) levels wereall determined. In addition, the expressions of the interleukin (IL)-8in lung and tumor necrosis factor (TNF)-α in heart were pathologically and immunohistochemically were determined.Results: Sepsis caused significant increases in CK, LDH, LPO, GSHand heart CAT activity and caused inhibition of SOD and lung CATactivity (p<0.001). The OLE significantly decreased the LPO and GSHlevels (p<0.001) and increased the SOD in activity of both tissuesand CAT activity in lung tissue (p<0.001). Moreover, CEFT significantly decreased the LPO level (p<0.01) and increased tissue SODand CAT activities in lung tissue (p<0.001). CK and LDH activities inserum showed marked decrease in OLE and CEFT groups (p<0.001).Conclusion: Antioxidant and a notable anti-inflammatory effect ofOLE by reducing IL-8 and TNF-α levels have been stated. Thus, astrong antioxidant, such as OLE, would be beneficial for the treatment of sepsis
